Acknowledgements Acknowledgements 2 First of all I would like to thank Prof. Marcus Groettrup for giving me the opportunity to perform my PhD in his group. Thank you for supplying optimal working conditions and providing scientific guidance over the past years. Thank you to Prof. Martin Scheffner's group for the collaboration in the UBE1L2 paper and for lots of advice. I would like to express my gratitude to Dr. Gunter Schmidtke, who helped me a lot with biochemical problems and didn't get tired of explaining things to me. Thank you for providing useful technical tips and tricks. Special thanks go to Ingrid Kassner who made my PhD years more fun. It was great to have you as a " Vertiefungskurs " student, a diploma student and as a " HiWi ". I also had a privilege to work with very nice and skilled people in the FAT10 lab. My thanks go to my fellow PhD students Birte, Sebastian and Neha. Further thanks go to Elisabeth and Tina who technically supported us in the lab. I very much appreciate the collaboration with Annette Aichem at the BITg. Thank you for giving me the opportunity to participate at the E2 story. Thank you also to the rest of the immunology lab members for the great atmosphere in our Furthermore, I would like to thank Carolina and Jacky who became good friends over the last years. Thank you for your support. Finally, special thanks go to my family and all my friends, who constantly supported me during my PhD. Thank you for always being there for me.

Conjugation of the Ubiquitin Activating Enzyme UBE1 with the Ubiquitin-Like Modifier FAT10 Targets It for Proteasomal Degradation

The ubiquitin-like modifier HLA-F adjacent transcript 10 (FAT10) directly targets its substrates for proteasomal degradation by becoming covalently attached via its C-terminal diglycine motif to internal lysine residues of its substrate proteins. The conjugation machinery consists of the bispecific E1 activating enzyme Ubiquitin-like modifier activating enzyme 6 (UBA6), the likewise bispecific ...

Ubiquitin-like Protein Conjugation: Structures, Chemistry, and Mechanism

Ubiquitin-like proteins (Ubl's) are conjugated to target proteins or lipids to regulate their activity, stability, subcellular localization, or macromolecular interactions. Similar to ubiquitin, conjugation is achieved through a cascade of activities that are catalyzed by E1 activating enzymes, E2 conjugating enzymes, and E3 ligases. In this review, we will summarize structural and mechanistic ...

Activating the ubiquitin family: UBA6 challenges the field.

Since its discovery in 1981, ubiquitin-activating enzyme 1 was thought to be the only E1-type enzyme responsible for ubiquitin activation. Recently, a relatively uncharacterized E1 enzyme, designated ubiquitin-like modifier activating enzyme 6, was also shown to activate ubiquitin. Ubiquitin-activating enzyme 1 and ubiquitin-like modifier activating enzyme 6 are both essential proteins, and eac...

USE1 is a bispecific conjugating enzyme for ubiquitin and FAT10, which FAT10ylates itself in cis.

The ubiquitin-like modifier FAT10 targets proteins for degradation by the proteasome and is activated by the E1 enzyme UBA6. In this study, we identify the UBA6-specific E2 enzyme (USE1) as an interaction partner of FAT10. Activated FAT10 can be transferred from UBA6 onto USE1 in vitro, and endogenous USE1 and FAT10 can be coimmunoprecipitated from intact cells. Small interfering RNA-mediated d...

A novel protein modification pathway related to the ubiquitin system.

Ubiquitin conjugation is known to target protein substrates primarily to degradation by the proteasome or via the endocytic route. Here we describe a novel protein modification pathway in yeast which mediates the conjugation of RUB1, a ubiquitin-like protein displaying 53% amino acid identity to ubiquitin. We show that RUB1 conjugation requires at least three proteins in vivo. ULA1 and UBA3 are...
